HELP! What is the best route through NY when I want to see Lion King, Statue of Liberty, Central park....?
Botanic Garden

Madison Square Garden, Times Square, Rockafella, Empire State, Brooklyn Bridge, and do some shopping

so that I don't waste time running around the same area twice?

I'll be there for about 4 days..

Please suggest some other MUST SEE attractions/restaurants/nightclubs...

Grouping things that are in the same general area:
Central Park is the most uptown thing you listed (110 Street down to 59 Street between Fifth Avenue and Central Park West)). From there, the next spot going downtown is Rockefeller Center (47 to 50 Street, between Fifth and Sixth Avenues). Then Times Square (42 to 46 Streets, Broadway and Seventh Avenue), Madison Square Garden (33-34 Streets at Eighth Avenue); Empire State Building (33-34 Streets on Fifth Avenue); the Brooklyn Bridge, and finally, the boat to Ellis Island and the Statue of Liberty (it's the same boat, one price, stops at Ellis first; you can get out at each to explore and take a later boat onward--warning, very LONG lines form so there can be a long wait to board, start early).
As fort Brooklyn Bridge, there is a subway stop near the entrance. You can walk across this bridge, it having been built before Robert Moses messed up the city. This subway stop is also right by the beginning of Chinatown.
Additional sights going from uptown to downtown:
Metropolitan Museum of Art and Museum of Natural History/Rose Space Center are on opposite sides of Central Park.
58 Street and Fifth Avenue is FAO Schwartz, giant toy store with an ice cream parlor.
First Avenue and 44 Street, the United Nations (gift shop is a great place for souvenirs). The side streets off Times Square have a number of very good restaurants.
Lower Manhattan near the boat for Ellis/Liberty; World Trade Center site; Fraunces Tavern (where Washington gave his farewell address); Staten Island Ferry (free, great view of harbor, one hour round trip); Chase Bank Money Museum; South Street Seaport; Chinatown; stock exchange.
Outside Manhattan: Brooklyn Aquarium; Coney Island
Bronx Zoo; Botanical Gardens
